High-Dose Riboflavin (Vitamin B2) for Migraine Prevention
Symptoms Studied
Recurring migraine headaches with throbbing pain, often one-sided, accompanied by nausea, photophobia, and phonophobia. Attacks occurring 2-6 times per month, significantly impacting quality of life.
Research Hypothesis
Riboflavin plays a vital role in mitochondrial energy production. Since migraine pathogenesis includes mitochondrial dysfunction, supplementation may reduce migraine frequency by improving cellular energy metabolism.
Intervention Tested
Riboflavin (Vitamin B2)
400mg riboflavin (vitamin B2) daily, taken orally as a single dose
Study Results
Before Treatment
Patients with migraine meeting IHS criteria, experiencing 2-6 attacks per month with significant functional impairment.
After Treatment
59% of riboflavin patients achieved 50% or greater reduction in migraine frequency compared to 15% for placebo (p<0.01). Significant reduction in migraine days and attack frequency.
Timeline to Improvement
Improvements typically observed by week 4-6, with continued improvement through 3 months
Side Effects
Generally well-tolerated. Bright yellow urine discoloration (harmless). Occasional mild GI discomfort.
